Abstract

Endocrine disruptors are exogenous chemical substances to the organism, capable of altering the homeostasis of the endocrine- reproductive system. A group of these, estrogenic compounds, may cause negative effects on reproductive capacity of some fish, decreasing fertility, sexual ability, sperm production and feminization of males. This paper describes the analytical performance of a method for the quantitation of estrogenicity in water. The technique used was the Yeast Estrogen Screen (YES) with recombinant Saccharomyces cerevisiae, a genetically modified yeast with human estrogen receptor expression and a plasmid with the lacZ reporter gene under the control of estrogen response elements. The gene coding for β-galactosidase, which is secreted to the medium catalyzes the metabolism of a chromogenic substrate, showing a color change measure to 420 nm. The standard 17 β-estradiol was used to build a calibration curve in the range of concentrations between 1.75 ng/l and 7.5 μg/l. Sigmoid curves of dose - response obtained were fitted by the Hill function and r 2 was greater than 0.95. The method detection limit is 55 ng/l. The analyzed samples showed no detectable estrogenicity.
